Results
Below are the results from a preliminary trial. The cells were transfected with GDPD6, not treated with estrogen, and tested to make sure expression levels were accurate. A western blot (Figures 2-4) was done to this end, as well as determining the fold change in mRNA expression (Figure 1) (a higher number means more expression and a lower number means a lower expression.
Analysis
Based on the results, we can conclude that the transfection did not work. In Figure 1, GDPD6 empty-vector cell lines are denoted by the blue bar, GDPD6 over-expression is denoted by the red bar, and GDPD6 wild-type is denoted by the green bar. In theory, if the transfection worked, the fold change in mRNA would be higher in the red bar than in the blue but that is not the case. This allows us to conclude the transfection did not work and will need to be repeated on a new cell line.
For the western blots, three trials were run for each cell line. The line thickness for the empty-vector and over-expressing cell lines were consistently the same in each trial. This means that in the cell lines, the empty-vector and over-expressing cell lines were expressing GDPD6 at the same levels. This further solidifies the conclusion that the transfection did not work.
